QuEChERS-Based Method for the Analysis of OPP (sum)

A QuEChERS-based method (AcH-QuEChERS) for the analysis of 2-phenylphenol (OPP) and its conjugates in food of plant origin.

The glycosidic linkage of OPP to sugars is chemically quite resistant to alkaline hydrolysis and also to weak acidic hydrolysis. Strong acidic hydrolysis conditions are required for de-conjugation: In the case of fruits and vegetables, 2 mL 10 N H2SO4 are added to 5g sample. The reaction takes place within a heated shaker at 70°C for 16-24 h.

Despite being less sensitive than GC-MS/MS for the analysis of OPP, LC-MS/MS is proposed for measurement as OPP interferences are frequently observed in GC-MS/MS.

For the direct analysis of OPP-Gucoside (OPP-Glc) satisfactory recoveries and good sensitivity is achieved using CEN-QuEChERS in combination with LC-MS/MS. Nevertheless, OPP (sum) cannot be simply calculated by adding up the results of OPP and OPP-Glc, as other types of conjugates form a considerable share of conjugated-OPP.
